Effects of Cyclobuxine D on Longitudinal Smooth Muscle Isolated from the Rabbit Jejunum

Abstract

Cyclobuxine D is a steroidal alkaloid, ivhich is extracted from Buxus microphylla var. koreana Nakai. Many pharmacological effects of cyclobuxine D was clarified in our Lab.: the anti-infammatory effe Vt in carrageenin-induced inflammation; the bradycardic effect and effects on electrocardiogram in rats.
In this experiment, we examined the effect of cyclobuxine D on the isolated longitudinal muscle from the rabbit jejunum. Cyclobuxine D showed a dose-dependently significant relaxing effect on the isolated rabbit jejunum. The increase in tension induced by acetylcholine (5.5 x 10-7M) was completely blocked by cyclobuxine D (1.5 x 10-¢¥M). Atropine (4.7 x 10-5M), a muscarinic antagonist, did not affect the relaxing effect of cyclobuxine D (1.5 x 10-4M). Cyclobuxine D (1.5 x 10-¡ÆM) blocked significantly the increase in tension elicited by barium chloride (4.1 x 10-5M) which can activate the contractile protein itself. Cyclobuxine D and papaverine (a direct smooth muscle relaxant) exerted a synergistic effect on the isolated longitudinal muscle from the rabbit jejunum.
